I read about the PI's lab on his website and asked via email if I could join his team as an undergraduate research assistant. He invited me for a 30 minute in person interview, during which we discussed what type of research he typically does and what my responsibilities would consist of, if I were to be hired.

Simple process. Enthusiasm, and interest is of utmost importance. I applied for a biology research assistant position. Your experience will largely depend on the research lab you join. Depending on group, some work experience is necessary. Class experience can sometimes substitute for experience, but you should do well in your classes!
